SiliaBond® Reagents Kit For Microwave Applications

In recent years, μ-wave synthesizers have taken organic chemistry by storm. Fast kinetics, higher yields, excellent purities, wide compatibility of solvents, and their applicability to a variety of reactions and applications, makes them very important tools in the laboratory. After their introduction, chemists started to use supported reagents for solution-phase synthesis. The polymer supported reagents commonly used, although very useful, have drawbacks in -wave synthesizers: swelling and heat stability. The high temperatures generated inside these synthesizers put stress on these resins. Also, because of the small reaction volumes, the swelling of the resins can be problematic. Silica-based reagents on the other hand, do not suffer from such shortcomings. They are heat resistant and they do not swell.

For your convenience, SiliCycle and CEM Corporation had developed a silica-based reagents kit for μ-wave applications that contains SiliaBond® reagents for different types of reactions (amide synthesis, reductive amination, Suzuki couplings) as well as electrophile, nucleophile, and metal scavenging reactions that demonstrate the effectiveness of these reagents in μ-wave applications. You will find general experimental procedure related to each application.

    SiliaBond® Amine (Si-NH2) is an effective scavenger of acid chlorides, sulfonyl chlorides, isocyanates and other electrophiles. Si-NH2 has been shown to be effective metal scavenger and catalyst for Knoevenagel reactions. SiliaBond Amine is also used in chromatography as normal phase sorbent.

    SiliaBond® Carbonate

    SiliaBond® Carbonate (Si-CO3) is the silica bound equivalent of tetramethyl ammonium carbonate. It can be used as a general base to quench a reaction, to free base amines in their ammonium salt form and to scavenge acids and acidic phenols, including HOBt, which is widely used in amide coupling reactions.

    SiliaBond® Cyanoborohydride

    SiliaBond® Cyanoborohydride (Si-CBH) is the silica bound equivalent of sodium cyanoborohydride. Bound cyanoborohydride is very useful in reductive amination and in the reduction of imines and aldehydes. This problem is minimized with the use of silica bound materials since the toxic cyanide residue remains on the silica.

    SiliaBond® Ethyl-Dimethylaminopropyl Carbodiimide (Si-EDC) reacts in a similar fashion to SiliaBond Carbodiimide (Si-DCC). The advantage of Si-EDC is this new reagent greatly simplifies the work up procedure of the reactions since a simple filtration is all that takes to remove the unwanted side product of the amides formation (urea).

    SiliaBond® Diethylamine (SiliaBond WAX-2) is a silica bound tertiary amine base and can be used in most applications requiring a tertiary amine, particularly as a HCl sponge. It is a very common supported tertiary amine.

    SiliaBond® TBD

    SiliaBond® TBD (1,5,7-Triazabicyclo[4.4.0]dec-5-ene, Si-TBD) is a silica bound bicyclic guanidine moiety that is sufficiently basic to deprotonate moderately acidic hydrogens. It is most commonly used for the alkylation of phenols (Williamson ether synthesis) and amines, and the esterification of carboxylic acids using alkyl halides. It may also be used to scavenge boronic acids such as phenylboronic acid: 98% removal with 1 equivalent in DCM at room temperature after 1 h.

    SiliaMetS® Thiourea (Si-THU) is a versatile metal scavenger for all forms of palladium and is widely used in the pharmaceutical industry. It works particularly well in organic solvents. It can also be used to scavenge Ag, Pt, Ru, Rh and Hg. Once complexed with a transition metal, it has been reported as being an effective catalyst.
