What is the appropriate flow rate for the different column dimensions?
Typical flow rates vs column id for optimal column efficiency vary according to the van Deemter equation.
Internal Dimension | 3 µm | 5 µm | 10 µm |
---|---|---|---|
2.1 mm ID | 0.3 mL/min | 0.21 mL/min | - |
3.0 mm ID | 0.60 mL/min | 0.43 mL/min | - |
4.6 mm ID | 1.50 mL/min | 1.00 mL/min | 0.7 mL/min |
10 mm ID | - | 4.70 mL/min | 7.7 mL/min |
21.2 mm ID | - | 21.0 mL/min | 18.0 mL/min |
30mm ID | - | 42.0 mL/min | 38.0 mL/min |
50 mm ID | - | - | 118.0 mL/min |
For increased lab productivity and efficiency and in order to increase sample throughput and to achieve shorter separation times, resolution permitting, the flow rate can be increased until the pressure limit of the HPLC pump is reached (6000 psi).
