How do you determine the quantity of silica required in a SPE tube?

The mass of the sample, rather than the volume of the sample, will be the factor to consider. Typically, the sorbent capacity will be around 5 % (weight/weight) for silica-based products and around 10 % (weight/weight) for polymeric products.

Silica-Based (SiliaPrep): load up to 5 % of bed weight.
e.g. 100 mg of silica-based sorbent will retain up to 5 mg of sample

Polymeric (SiliaPrepX): load up to 10 % of bed weight.
e.g. 100 mg of polymeric sorbent will retain up to 10 mg of sample

Tips

  • Not enough sorbent: ANALYTE LOSS ► low recovery and reproducibility
  • Too much sorbent: MORE EXPENSIVE ► more solvent used, taller SPE cartridges
  • Concentrated samples: double the bed weight to avoid analyte loss
