Sample Pretreatment before Loading
Add 40 mL of Methanol / Water (80:20) and 0.2 g of Sodium Chloride to 5 g of peanut butter. Stir for 2 hours, filter on paper and rinse with 15 mL of Methanol. Dry the combined extracts with Magnesium Sulfate and evaporate under Nitrogen. Reconstitute with 500 μL of Methanol / Water (80:20).

Washing 2 (Silica)
3 x 2 mL of Methanol / Chloroform (50:50).
Further Treatment 2 (Silica)
Combine loading and washing fractions, evaporate under Nitrogen, reconstitute with 0.1 % Formic Acid and Ammonium Acetate (5 mM) in Water / Methanol (50:50) and quantify by LC/MS.
