Sample Pretreatment before Loading

Weigh 5 g of mixed eggs or frozen and milled meat samples, or 10 mL of bovine milk.
Add 20 mL of the extraction solution: NH4OAc (10 mM), EDTA (0.4 mM), NaCl (0.5 %) and Trichloroacetic Acid (2 %) in Water.
Vortex and centrifugate at 4,000 rpm for 10 minutes.
Transfer the supernatant to a clean tube.
Repeat extraction and combine supernatants.
Adjust pH to 6.5.

10 mL of Methanol

Equilibration

10 mL of Water

Loading

Pretreated sample

Washing

10 mL of Ammonium Acetate buffer (50 mM, pH 7.0)
Dry the cartridge

Elution

6 mL of 2 % Formic Acid in Water

Further Treatment

Dilute with 4 mL of Water, filter on 0.2 μm and quantification by LC/MS/MS.
