Sample Pretreatment before Loading
Add 3 mL of Phosphate buffer (100 mM, pH 6.0) to 2 mL of blood / plasma / serum / urine (or 1 g of tissue homogenate).
Mix / vortex.
Adjust pH to 6.0 ± 0.5 with Sodium Phosphate (100 mM).
Centrifugate at 2,000 rpm for 10 minutes and discard cellular debris.
Conditionning
3 mL of Methanol
Equilibration
5 mL of Phosphate buffer (100 mM, pH 7.0)
Loading
Pretreated sample
Dry the cartridge
Washing
No washing required
Elution
6 mL of 1 % Ammonium Hydroxide in Methanol
Further Treatment
Evaporation down to 1 mL under Nitrogen and quantification by LC/MS/MS.
