Scale-Up of a Polonovski Reaction – Impact of Residual Palladium in Starting Material

For the kilogram-scale manufacturing of MCL-509 Amine·HCl, a potential prodrug for Parkinson’s disease, the synthesis pathway had to be re-examined for an easier, safer, and more efficient scale-up.

A Metal Scavenger treatment was performed using SiliaMetS Thiol. In doing so, the Pd content was reduced below the quantification limit of the ICP analysis which was considered acceptable and allowed better control and optimization of the kilogram-scale N-methylation of MCL-509 Amine·HCl.
